Το WD40 πρωτεϊνικό μοτίβο αποτελεί ένα από τα πιο ευρέως διαδεδομένα μοτίβα του πρωτεώματος των ευκαρυωτικών οργανισμών. Η ικανότητα των πρωτεϊνών που περιέχουν WD40 επαναλήψεις (WDR πρωτεΐνες) να αναδιπλώνονται σε τριτοταγή δομή β-προπέλας, τις καθιστά ιδανικά ικριώματα για πρωτεϊνικές αλληλεπιδράσεις. Στα φυτά, οι WDR φαίνεται να αποτελούν κεντρικοί ρυθμιστές μίας πληθώρας κυτταρικών και αναπτυξιακών διεργασιών. Το πρωτέωμα του Arabidopsis thaliana περιέχει μία πολυπληθή υπεροικογένεια WDR πρωτεϊνών, η οποία σύμφωνα με τελευταίες εκτιμήσεις αριθμοί περισσότερα από 400 μέλη (Sharma and Pandey, 2016). Το γονίδιο εδράζεται στο πέμπτο χρωμόσωμα του Arabidopsis, στο γενετικό τόπο 66240 και η πρωτεΐνη που παράγει αποτελείται από 331 αμινοξέα. Σύμφωνα με τη συγκριτική μελέτη των Lee και συνεργατών του (2008) αποτελεί πιθανή υπομονάδα ενός Ε3 Cullin-RING συμπλόκου λιγάσης, ενώ εμφανίζει μεγάλο ποσοστό αμινοξικής ταυτότητας με μία ομόλογη πρωτεΐνη στο πρωτέωμα του Arabidospsis, την AtAPRF1. Παρά τη μεγάλη ομοιότητάς τους, δεδομένα από τη λειτουργική ανάλυση των δύο πρωτεϊνών (παρούσα μελέτη και αδημοσίευτα αποτελέσματα) υποδεικνύουν ότι συμμετέχουν σε δύο διακριτά σύμπλοκα. Η AtULCS1, όπως αποκαλύπτουν τα αποτελέσματα της συγκεκριμένης εργασίας, αλληλεπιδρά in planta με την DDB1a υπομονάδα, δημιουργώντας ένα λειτουργικό σύμπλοκο CUL4-DDB1aAtULCS1 του πρωτεολυτικού μονοπατιού, ρυθμίζοντας βασικές κυτταρικές διεργασίες. Αντίθετα, η ομόλογη AtAPRF1, εμπλέκεται στη ρύθμιση του χρόνου άνθησης στο Arabidopsis, πιθανά μέσω μεθυλίωσης του γενετικού τόπου FLC.Κατά την πειραματική προσέγγιση της λειτουργικής ανάλυσης του γονιδίου, πραγματοποιήθηκε η in silico ανάλυση του AtULCS1 όσο και της πρωτεΐνης που κωδικοποιεί, ενώ ταυτόχρονα μελετήθηκε και η φυλογένεση της. Από την ανάλυση αυτή, διαπιστώθηκε η ύπαρξη δύο ομόλογων αλληλουχιών σε όλους τους φυτικούς οργανισμούς που ελέγχθηκαν. Αυτές οργανώνονται σε δύο διακριτούς κλάδους, διαφορετικούς από αυτόν των μυκήτων και των ζώων. Η πρωτεΐνη AtULCS1 εντοπίζεται τόσο στον πυρήνα όσο και στο κυτταρόπλασμα των κυττάρων, όπως αποκαλύπτουν τα αποτελέσματα του υποκυτταρικού εντοπισμού της, ενώ αλληλεπιδρά in planta ειδικά με την DDB1a υπομονάδα του συμπλόκου Ε3 λιγάσης. Η μελέτη του ιστοειδικού προτύπου του γονιδίου, μέσω της δημιουργίας διαγονιδιακών σειρών μεταγραφικής σύντηξης του υποκινητή με το γονίδιο αναφοράς της β-γλυκουρονιδάσης (GUS), υπέδειξε την έκφραση του σε πολλά αναπτυξιακά στάδια, με μέγιστη έκφραση στους ιστούς του άνθους. Επιπρόσθετα, δημιουργήθηκαν και αναλύθηκαν σε μοριακό και φαινοτυπικό επίπεδο διαγονιδιακές σειρές RNAi αποσιώπησης, καθώς και υπερέκφρασης του γονιδίου, οι οποίες αποκάλυψαν την εμπλοκή του AtULCS1 γονιδίου σε μία πληθώρα αναπτυξιακών διεργασιών, με σημαντικότερη την εναπόθεση δευτερογενών τοιχωμάτων στους ιστούς του ενδοθηκίου και του αγωγού ιστού του βλαστού. Τέλος, όσον αφορά το μοριακό μονοπάτι στο οποίο συμμετέχει, τα δεδομένα υποστηρίζουν ότι εμπλέκεται στη ρύθμιση βασικών ΜΥΒ μεταγραφικών παραγόντων, οι οποίοι ελέγχουν την ενεργοποίηση ή καταστολή γονιδίων των βιοσυνθετικών μονοπατιών της λιγνίνης και κυτταρίνης.

However, the biological signi cance of most of these proteins is still unknown, while only a few of them have associated biological functions.The current PhD thesis aimed in characterizing the AtULCS1 (Ubiquitin Ligase Complex Subunit 1) gene from Arabidopsis thaliana, which encodes for a novel low molecular weight WDR protein. AtULCS1 consists of 331 amino acids and contains ve WD40 repeats that form the structural WD40 region of the protein. Based on a whole-genome in silico analysis from Arabidopsis, AtULCS1 was annotated as a putative subunit of an E3 Cullin-Ring Ligase (CRL) complex.The phylogenetic analysisrevealed the existence of two homologous proteins in all plant species, which are organized in two distinct clades of the phylogenetic tree. In the Arabidopsis proteome, the two homologous proteins are AtULCS1 and AtAPRF1, which share a high percentage of amino acid identity. However, based on our data (current PhD thesis and unpublished data) the two proteins appear to be subunits of distinct complexes and exhibit di erent molecular function. supports the involvement of the two proteins in distinct complexes. AtULCS1 interacts in planta with DDB1a, the protein subunit of DWD E3 CRL complexes, which regulate a variety of cellular and developmental processes through the proteolytic machinery. On the other hand, AtAPRF1 is suggested to control owering time in Arabidopsis, possibly through epigenetic regulation of the f FLC genetic locus (Kapolas et al., 2016).To obtain insights into the tissue speci c expression pattern of AtULCS1, transgenic Arabidopsis plants harboring a promoter-GUS transcriptional fusion construct were analyzed. The analysis revealed that AtULCS1 is expressed during development in various plant tissues, while the maximum expression was found to be in oral tissues. The localization of AtULCS1 in both the nucleus and cytoplasm in Nicotiana benthamiana epidermal cells implies the possible assembly of distict E3 CRL complexes, functionally associated with these cellular compartments.In order to unravel the role of AtULCS1 in plant development, ann RNAi-mediated silencing and overexpression approach was used. Detailed phenotypic analysis of the ulcs1i transgenic lines revealed the involvement of the protein in a variety of developmental processes, such as seed size regulation, cell proliferation and di erentiation, plant growth and secondary wall modi cation. The inability of ulcs1i mutants to deposit cellulose and lignin in anther endothecium cells, leads to a sterile phenotype due to anther indehiscence. Moreover, ulcs1i plants exhibit less cells with secondary wall thickening in the vascular tissue of the shoot, indicating a functional role of the gene during secondary growth. On the contrary, overexpression of AtULCS1 leads to a reversal of the phenotype, con rming its importance in secondary wall modi cations. Our molecular data suggest that AtULCS1 participates in complexes, which regulate the expression of several MYB transcription factors during secondary growth. Deregulation of these “master switches” leads to a disturbance of the expression of several IRX biosynthetic genes and nally to an impairment in secondary wall modi cations of endothecium and vascular xylem cells.
