Στα πλαίσια της παρούσας διδακτορικής διατριβής πραγματοποιήθηκε μια διεπιστημονική ερευνητική εργασία, η οποία είχε ως κεντρική ιδέα τη χημεία και τη βιοδραστικότητα των οργανοκασσιτερικών ενώσεων. Για την επίτευξη της μελέτης αυτής, η ερευνητική εργασία κινήθηκε σε τρεις κεντρικούς άξονες: σχεδιασμός και σύνθεση, ανάλυση και ταυτοποίηση και, τέλος, in vitro μελέτη βιολογικών δράσεων. Αρχικά σχεδιάστηκαν και συντέθηκαν ιμίνες (βάσεις Schiff) παράγωγα του βενζοϊκού οξέος, από τη συμπύκνωση του 3- και του 4-αμινοβενζοϊκου οξέος με π-υποκατεστημένες (Η, ΝΟ2, Cl, MeO, Me2N) βενζαλδεΰδες, καθώς και από τη συμπύκνωση των π-υποκατεστημένων (Η, ΝΟ2, Cl, MeO, Me2N) ανιλίνων με π-φορμυλοβενζοϊκό οξύ. Από τις αντιδράσεις αυτές συντέθηκαν βιολογικά ενεργά αρυλιμηνο υποκατεστημένα βενζοϊκά οξέα, τα οποία χρησιμοποιήθηκαν στη συνέχεια της συνθετικής διαδικασίας ως ligands (L) για τη σύνθεση νέων τριοργανοκασσιτερικών εστέρων γενικού τύπου R3SnL με R= Μe και Ph. Στα πλαίσια της παρούσας διδακτορικής διατριβής πραγματοποιήθηκε μια διεπιστημονική ερευνητική εργασία, η οποία είχε ως κεντρική ιδέα τη χημεία και τη βιοδραστικότητα των οργανοκασσιτερικών ενώσεων. Για την επίτευξη της μελέτης αυτής, η ερευνητική εργασία κινήθηκε σε τρεις κεντρικούς άξονες: σχεδιασμός και σύνθεση, ανάλυση και ταυτοποίηση και, τέλος, in vitro μελέτη βιολογικών δράσεων. Αρχικά σχεδιάστηκαν και συντέθηκαν ιμίνες (βάσεις Schiff) παράγωγα του βενζοϊκού οξέος, από τη συμπύκνωση του 3- και του 4-αμινοβενζοϊκου οξέος με π-υποκατεστημένες (Η, ΝΟ2, Cl, MeO, Me2N) βενζαλδεΰδες, καθώς και από τη συμπύκνωση των π-υποκατεστημένων (Η, ΝΟ2, Cl, MeO, Me2N) ανιλίνων με π-φορμυλοβενζοϊκό οξύ. Από τις αντιδράσεις αυτές συντέθηκαν βιολογικά ενεργά αρυλιμηνο υποκατεστημένα βενζοϊκά οξέα, τα οποία χρησιμοποιήθηκαν στη συνέχεια της συνθετικής διαδικασίας ως ligands (L) για τη σύνθεση νέων τριοργανοκασσιτερικών εστέρων γενικού τύπου R3SnL με R= Μe και Ph. Η ταυτοποίηση και ο χαρακτηρισμός των νέων αυτών ligands και των συμπλόκων που συντέθηκαν πραγματοποιήθηκε με συνδυασμό αναλυτικών τεχνικών: φασματοσκοπικές τεχνικές (FT-IR, 1H-NMR, 13C-NMR, UV-vis), στοιχειακή ανάλυση και την κρυσταλλογραφία ακτίνων Χ. Επόμενο στάδιο ήταν η in vitro μελέτη βιοδραστικότητας των ενώσεων αυτών η οποία, χωρίστηκε σε δύο σκέλη. Το ένα βιολογικό σκέλος κινήθηκε σε κυτταρικό επίπεδο, όπου αξιολογήθηκε πρώτα η in vitro αποπτωτική δράση σε τέσσερεις καρκινικές κυτταρικές σειρές, ΗepG2, HeLa, K562 και N2a. Για το σκοπό αυτό χρησιμοποιήθηκε η δοκιμή ΜΤΤ (ΜΤΤ assay), η οποία εκτιμά την επίδραση στη βιωσιμότητα των κυττάρων μετά από έκθεσή τους σε διάφορες συγκεντρώσεις των ουσιών. Στη συνέχεια έγιναν νευροκυτταροτοξικές μελέτες, εκτιμώντας την ενδεχόμενη in vitro αναπτυξιακή νευροτοξικότητα. Για το σκοπό αυτό αξιολογήθηκε η επίδραση των υπό μελέτη ουσιών στη διαδικασία ανάπτυξης αξονικών προεκβολών σε υπό διαφοροποίηση Ν2a κύτταρα, μετά από έκθεση σε υποκυτταροτοξικές συγκεντρώσεις των υπό μελέτη ουσιών. Το άλλο βιολογικό σκέλος κινήθηκε σε μοριακό επίπεδο, στοχεύοντας στην εκτίμηση της επίδρασης των υπό μελέτη ουσιών σε συγκεκριμένους βιοχημικούς παράγοντες με σκοπό τη διαλεύκανση των βιοχημικών μηχανισμών δράσεών τους. Εκτιμήθηκε η δράση τους σε συγκεκριμένες πρωτεΐνες του νευροκυτταρικού σκελετού, χρησιμοποιώντας SDS-PAGE και ανοσοχημική τεχνική ανάλυσης Western blot. Επιπλέον, έγινε εκτίμηση της in vitro ανασταλτικής δράσης τους στη λιποξυγονάση, ένζυμο που είναι γνωστό για το ρόλο του στην παραγωγή ελευθέρων ριζών στη φλεγμονή. Τέλος, εκτιμήθηκε η ενδεχόμενη in vitro αντιοξειδωτική δράση τους, αξιολογώντας την ικανότητά τους στην αναστολή της επαγόμενης από το ΑΑΡΗ λιπιδικής υπεροξείδωσης, καθώς και την ικανότητα αλληλεπίδρασής τους με τη σταθερή ρίζα DPPH. Από τα αποτελέσματα που προέκυψαν προσπαθήσαμε να βγάλουμε συμπεράσματα για το κατά πόσο η δομή των καρβοξυλικών οξέων που χρησιμοποιήθηκαν ως Ligands επηρεάζει τόσο τον τρόπο συναρμογής της καρβοξυλικής ομάδας με το τριοργανοκασσιτερικό κατιόν, όσο και τη βιολογική δράση των υπό μελέτη τριοργανοκασσιτερικών εστέρων στις προαναφερόμενες μελέτες.

Ιn a subsequent step these compounds were used as ligands, HL, for the preparation of triorganotin derivatives of the general formula SnR3L. The reaction is a simple metathesis one, carried out in a suitable solvent in order to facilitate separation of KCl since triorganotin chlorides and the potassium salts of the above ligands were used as reactants. Unfortunately the tributyltin derivatives were prepared in the form of oils and we did not isolate pure solid compounds from them, limiting our study to the trimethyl and triphenyltin derivatives of the above substituted benzoates.The compounds were again characterized by a number of spectroscopic techniques (FT-IR, 1H-NMR, 13C-NMR), and for several of them X-ray diffraction was applied to serve for crystal structure determination. In accordance with the indications from the IR spectra and with the literature data available, the structural information verifies the electrophilic character of the tin centre which is coordinated to five atoms, three carbons from the organic substituents and two oxygen atoms. In the phenyl derivatives the second oxygen atom is asymmetrically coordinated from the same ligand giving rise to a chelate coordination and an asymmetrical overall tetrahedral tin environment, while in the methyl derivatives the second oxygen comes from a neighboring unit proceeding in the formation of long 1D polymeric structures. The above structures are not retained in solution as the NMR spectra have shown since there, evidence is that the tin environment is tetrahedral four-coordinate.The next step involved the bioreactivity study of some of the compounds which was partitioned in two directions. One was at a cell level, involving evaluation of the in vitro apoptotic action towards four cell lines, namely ΗepG2, HeLa, K562 and N2a. The evaluation of the studied compounds was carried out by means of the MTT assay for which a variety of compounds concentrations was applied to the cultures. Inspection of the calculated IC50 values gives some important information about the relative activity of the compounds. The apparent observation is that K562 are the more sensitive cell line being affected at low concentrations (below 0.5 μM) by nearly all the studied compounds. On the contrary the HeLa line is the more resistant with only one compound possessing IC50 value lower than 1 μΜ. The effect on line HepG2 cells lies in between the above extremes and some interesting variation of the observed values is made, namely that derivatives of the 4-imino substituted benzoic acid are more effective against the cells relative to their 3-imino counterparts. This variation indicates that in cases where there is no drastic action against the target cell, the electronic distribution within the triphenyltin carboxylate plays some role in determining the relative apoptosis procedure initiated by the compound. The above selectivity is interesting especially if it is taken into consideration that the best overall performance is presented by the MeO substituted 3-iminobenzoate, which has the best activity against both K562 and HeLa lines while for the HepG2 line it follows the pattern described above and presents only moderate action. However no simple relationship can be drawn with obvious or known structural parameters of the compounds and further investigations are required in this direction.Developmental neurotoxicity studies were performed by evaluating the non-toxic concentrations of the trimethyltin compounds studied towards N2a cells. In the first stage the three NO2-substituted ligands the 4-imino unsubstituted and the 4-imino NMe2 substituted one were tested in comparison with the well-characterized trimethyltin chloride (TMT). The NMe2 substitute derivative actually promoted the production of axial filaments while the rest four were found to be more active than TMT in inhibiting this process. In a subsequent step the compounds studied were tested by Western blotting for their effect towards several proteins involved in the cytoskeleton formation and function, namely total levels of NF-H, the phosphorylated NF-H, GAP43, α-tubulin, β-tubulin and HSP70. None of the compounds revealed uniform effect over all the studied proteins and none of the compounds revealed bioactivity similar to another, indicating that even if the main cause for the neurotoxicity effects is centered on the trimethyltin moiety, the structure of the benzoate plays a role in determining not only the extnent but also the path through which each compound acts.Interestingly enough the results relative to lipid peroxidation and lipoxygenase inhibition parallel those of the apoptotic behavior, indicating a redox pathway or a redox centered mechanism for the apoptosis activity of the triphenyl compounds. The structure of the benzoate plays a determinant role in the reactivity of those triphenyltin derivatives as either lipid peroxidation inhibitors or lipoxygenase inhibitors. Lipoxygenase inhibitory activity enhences when the aromatic ring substituent of the benzoic acid contains a nucleophilic group, while the lipid peroxidation inhibitory activity enhances in the presence of an electrophilic group. Lipid peroxidation is a process related to the formation of free radicals and in this respect the bioreactivity study related to the ability of free radical scavenging of the compounds as well as of their potential as inhibitors or key redox reactions within the cell was carried out. The studies were carried out in vitro and included AAPH induced peroxidation and investigation of the interaction with the stable free radical DPPH, however no significant action was detected owing to the fact of the sterically hindered nature of the lone electron in DPPH and the relative protection of the electrophilic tin centre by the organic substituents.
