Periodontal ligament cells concern a mixed heterogeneous cellular population that consists mainly of fibroblasts of mesenchymal origin. Subpopulations of those fibroblasts present osteoblastic characteristics and have the capacity to proliferate, migrate and produce more differentiated cells that can synthesize bone, cementum and the extracellular matrix of the periodontal ligament. Since the above mentioned processes are the bases of the remodeling and healing in the periodontal tissues, the study of the growth factors and hormones that may activate the biological mechanisms which promote the proliferation, the chemotaxis and the differentiation of the periodontal ligament cells is of great importance. The purpose of this study concerns the characterization of cells isolated from the human periodontal ligament was identified according to their mesenchymal origin and their fibroblastic and osteoblastic phenotype as well as the in vitro study of the effect of17-β estradiol and TGF-β1 on their proliferation. Three healthy premolars were extracted for orthodontic reasons from three adults and cells were isolated from their periodontal ligament. The mesenchymal origin of these cells was identified immunohistochemically by the detection of vimentin, their fibroblastic phenotype was estimated based on their morphology and the immunohistochemical detection of alpha-smooth muscle actin while their osteoblastic phenotype was identified on day four by measuring the alkaline phosphatase activity and by the quantitative determination of osteocalcin. Furthermore, the effect of 1ng/mL and 25ng/mL TGF-β1 ,as well as the effect of 1nM and 100nM 17-β estradiol on the proliferation of human periodontal ligament cells were tested at 48 and 72 hours. Cellular proliferation was estimated by counting the number of viable cells with the Neubauer hematocytometer after colorization of the cells with Trypan blue. Student’s t-test (p-value<0.05) for independent samples was applied for the comparison of the cell number within the groups between 48 and 72 hours while one way ANOVA(p-value<0.05) was performed for the comparison of the cell number between groups at 48 and 72 hours. The results of this study show that a)cells isolated from the human periodontal ligament are fibroblasts of mesenchymal origin that express osteoblastic phenotype, b) the transforming growth factor-beta1(TGF-β1) increases the proliferation of human periodontal ligament cells in a dose- and time-dependent manner, and the17-β estradiol increases, as well, the cellular proliferation in a dose-dependent manner.Περίληψη σε άλλη γλώσσα
Τα κύτταρα του περιρριζίου αφορούν ένα μεικτό ετερογενή πληθυσμό αποτελούμενο κυρίως από ινοβλάστες μεσεγχυματικής προέλευσης. Υποπληθυσμοί αυτών των ινοβλαστών φέρουν οστεοβλαστικό χαρακτήρα, μπορούν να πολλαπλασιαστούν, να μεταναστεύσουν και να παράγουν περισσότερο διαφοροποιημένα κύτταρα ικανά να συνθέσουν οστούν, οστεΐνη, και εξωκυττάρια θεμέλια ουσία. Ιδιαίτερο ενδιαφέρον παρουσιάζει η μελέτη των αυξητικών παραγόντων και ορμονών που συμβάλλουν στην ενεργοποίηση των βιολογικών μηχανισμών, οι οποίοι προάγουν τον πολλαπλασιασμό, τη χημειοταξία και τη διαφοροποίηση των περιρριζικών κυττάρων καθώς οι προαναφερθείσες διαδικασίες αποτελούν τη βάση για την επούλωση και ανάπλαση των περιοδοντικών ιστών. Η ορθή ερμηνεία, ωστόσο, των αποτελεσμάτων που προκύπτουν από αυτές τις μελέτες συχνά καθίσταται δύσκολη λόγω της ετερογένειας των κυτταρικών πληθυσμών που μπορούν να απομονωθούν από το ανθρώπινο περιρρίζιο. γεγονός αυτό καθιστά απαραίτητο το χαρακτηρισμό των κυττάρων αυτών πριν από τη χ ...
