Monocytes, mainly lymphocytes, were isolated by the Ficoll-Hypaque system and cultivaled in RPMI-1640. The immune response of the lymphocytes was studied by adding in the culture medium hydatid fluid and measuring the [³H] thymidine incorporation into the DNA of the cells. The viability of scolices and lymphocytes was determined by the Trypan Blue exclusion technique. All experiments were carried out in triplicate. The findings of the study demonstrate: 1. Lymphocytes from patients with echinococcosis are stimulated when cultured in the presence of hydatid fluid. Maximum stimulation occurred on the sixth day of culture, at a hydatid fluid protein concentration of 15 μg per ml of culture medium. The stimulation Index (SI) of the lymphocytes obtained from 10 subjects with active or cured echinococcosis was 4.3 ± 1.6 (mean ± SD), with a range from 7.0 to 2.1. Lymphocytes from normal control subjects demonstrated a SI of 1.1 ± 0.3, with a range from 0.5 to 1.5. 2. The immune memory of the sensitized lymphocytes is maintained at least up to 20 years after removal of the hydatid cyst, even if the Casoni's skin test and serology have become negative. 3. The viability of E. granulosus scolices cultivated in the presence of sensitized lymphocytes was significantly lower than that of the same scolices cultivated in the presence of nonsensitized lymphocytes. On the 5th day of culture only 5% of the scolices cocultivated with sensitized lymphocytes were alive, whereas on the same day of culture 57%, of the same scolices cocultlvated under similar conditions with nonsensitized lymphocytes were alive. 4. Sensitized lymphocytes were killed by E. granulosus scolices to a much greater extend than nonsensitized lymphocytes. 5. The reduced viability of both sensitized lymphocytes and scolices, when cocultivated, was not the result of a nutrient depletion, but was caused by the lymphocyte immune response. 6. The findings of this study indicate the presence long-lasting cellular immunity in echinococcosis and suggest that the production of a vaccine against E. granulosus is feasible.</dc:description><dc:description xmlns:lang="el">Δεδομένου ότι η ανοσολογική μνήμη μπορεί να παραμένει δια βίου, ακόμη και όταν δεν ανιχνεύονται αντισώματα στο ορό του ασθενούς, μελετήσαμε τη διέγερση λεμφοκυττάρων ατόμων, τα οποία έπασχαν ή είχαν νοσήσει από εχινόκοκκο μετά επώαση με υγρό υδάτιδος κύστεως. Τα αποτελέσματα της μελέτης αυτής δείχνουν την ύπαρξη κυτταρικής ανοσίας στην εχινοκοκκίαση και υποδηλώνουν τη δυνατότητα αναπτύξεως εμβολίου κατά της εχινοκοκκιάσεως.</dc:description><dc:title xmlns:lang="el">Καλλιέργεια λεμφοκυττάρων ευαισθητοποιημένων στον εχινόκοκκο ατόμων παρουσία προϊόντων υδατιδικής κύστεως: Βλαστική μεταμόρφωση λεμφοκυττάρων και δράση επί των σκωληκοκεφαλών</dc:title><dc:title xmlns:lang="en">Blast transformation of sensitized lymphocytes from patients with echinococcosis and killing of echinococcus ganulosus scolices by sensitized lymphocytes</dc:title><dc:creator xmlns:lang="en">Moustogianni-Kallimani, Anastasia</dc:creator><dc:creator xmlns:lang="el">Μουστόγιαννη-Καλλιμάνη, Αναστασία</dc:creator><dc:date>1990</dc:date><dc:language>gre</dc:language><dc:subject xmlns:lang="el">Βλαστική μεταμόρφωση</dc:subject><dc:subject xmlns:lang="el">Ευαισθητοποιημένα λεμφοκύτταρα</dc:subject><dc:subject xmlns:lang="el">Εχινοκοκκικό αντιγόνο</dc:subject><dc:subject xmlns:lang="el">Εχινόκοκκος granulosus</dc:subject><dc:subject xmlns:lang="el">Καλλιέργειες</dc:subject><dc:subject xmlns:lang="el">Κυτταρική ανοσία</dc:subject><dc:subject xmlns:lang="el">Λεμφοκυτταρική διέγερση</dc:subject><dc:subject xmlns:lang="el">Λεμφοκυτταρική μνήμη</dc:subject><dc:subject xmlns:lang="el">Πρωτοσκώληκες</dc:subject><dc:subject xmlns:lang="el">Υδατίδα κύστη</dc:subject><dc:subject xmlns:lang="en">Blast transformation</dc:subject><dc:subject xmlns:lang="en">Cellular immunity</dc:subject><dc:subject xmlns:lang="en">Culture</dc:subject><dc:subject xmlns:lang="en">Echinococcus granulosus</dc:subject><dc:subject xmlns:lang="en">Hydatid fluid</dc:subject><dc:subject xmlns:lang="en">Hydatid fluid's antigen</dc:subject><dc:subject xmlns:lang="en">Immune memory</dc:subject><dc:subject xmlns:lang="en">Lymphocytes stimulation</dc:subject><dc:subject xmlns:lang="en">Protoscolices</dc:subject><dc:subject xmlns:lang="en">Sensitized lymphocytes</dc:subject><dc:publisher xmlns:lang="en">University of Patras</dc:publisher><dc:publisher xmlns:lang="el">Πανεπιστήμιο Πατρών</dc:publisher><dc:subject xmlns:lang="el">Ιατρική και Επιστήμες Υγείας</dc:subject><dc:subject xmlns:lang="el">Κλινική Ιατρική</dc:subject><dc:subject xmlns:lang="en">Medical and Health Sciences</dc:subject><dc:subject xmlns:lang="en">Clinical Medicine</dc:subject><dc:identifier>10.12681/eadd/1497</dc:identifier><dc:identifier>http://hdl.handle.net/10442/hedi/1497</dc:identifier><dc:type>PhD Thesis</dc:type></oai_dc:dc></metadata></record></GetRecord></OAI-PMH>
